The 2D Membrane service is the extension of 2-Dimensional Differential In-Gel Electrophoresis (2D DIGE). This service includes trans-blotting the proteins from a 2-D gel to PVDF or Nitrocellulose membrane, and scanning the membrane images after the transfer. The membrane can be shipped back to scientists for them to perform Western blot, alternatively, the customers can provide primary antibodies and Applied Biomics will perform Western blot. These new services make the 2D Western blot feasible and convenient. There are mulitple advantages of the 2-D membrane service:
1) Visualizing 2-D protein images from both the 2-D gel and the PVDF membrane (before Western blot) without staining and destaining;
2) Imaging the 2-D membrane before Western blotting enables accurate matching of protein spots to the Western blot results;
3) Better sensitivity than other gel staining methods (Sypro Ruby, Deep purple, Lumitein and Flamingo staining) for the 2-D gel; superior sensitivity than other membrane staining methods (Poncesu S and Coomassie blue, etc)
4) Re-using the same membrane to detect multiple antigens by 2-D Western blotting when in combination with membrane stripping.
These new services combined with upstream 2D DIGE will allow scientists to visualize and identify proteins and their isoforms before and after post-translational modification including phosphorylation, glycosylation, ubiquitination, sulfation, deamidation, succination, etc. In addition, it can be used to validate the upstream 2-D DIGE data using the same technology platform, allowing for maximum reproducibility.
